Is it possible to run a REXX program in batch which has macro in it. I tried this but it did not work

Yes, it's done all the time. Don't forget that an ISPF Edit Macro requires ISPF, so ISPF has to be started and available, along with all of its necessary dataset allocations.

See 2.14 ISPF services in batch mode for more details.

I usually find it easier, if the intent is batch processing, to use a simple TSO EDIT instead. Depends on what you need to do.
